How to make a personal discount amount ?

I can think of two possible ways to do this - one method your customer can use, and one that you can...

 

it is entirely possible to setup a promotion code that can be used only once - after your client has used it during his purchase, it will no longer be valid for anyone else to use.

 

I know you don't seem to want to go down the promo code route - but it's probably the easiest way... you could send him a link to the product with the promo code added to it, so he wouldn't have to enter it himself.

 

another way would be for you to create the order on the admin side and use a price override - which would allow you to set the invoice at $200, but wouldn't mention the $50 discount they were receiving.

 

there would be other solutions too... e.g., you could just apply a $50 credit to the account and then reissue the invoice... but these occur after the purchase has been made, and I assume you want to do this before the invoice is issued.
